Write a Short Note on Vitali Test

The Vitali test or Vitali-Morin test is a specific method for detecting tropane alkaloids in plant materials. The test is named after two scientists, Vitali and Morin, who developed it. 

Vitali Test Principle

When atropine is treated with sulphuric acid followed by evaporation to dryness on a water-bath, it forms a polynitrocompound in yellow color. The yellow-colored compound dissolved in acetone on treatment with freshly prepared 0.5 ml of a 3% w/v solution of KOH in methanol shows a violet-red color that slowly changes to red. 

The procedure for the Vitali-Morin test typically involves the following steps:

  • A small amount of the plant material or its extract is dissolved in a suitable solvent, usually methanol or ethanol.
  • Add a few drops of concentrated smoky sulfuric acid into the test solution.
  • The color change can range from yellow to orange, red, violet, or blue, depending on the specific alkaloid present.

The Vitali Morin test is also conducted to identify flavonoids by treating the plant extract with concentrated sulfuric acid and observing the development of a green fluorescence under UV light.
